The stepwise coupling efficiencies
of the minor nucleotides are generally less than the efficiencies
we guarantee for normal DNA bases (G,A,T,C). As a consequence,
the yield of full length oligo may be reduced relative to an oligo
that does not contain a minor nucleotide. While we lot-select components
to obtain the highest possible coupling efficiencies and yields
of these minor nucleotides, the gel photos may show less than full
length bands corresponding to the positions of the minor nucleotides
in the sequence.
All oligos are synthesized
from the 3' end on solid supports. Selecting a modified 3' end
is not recommended, particularly at the jumbo synthesis scale, as
this may reduce quality and yield.
